NEWS: TdB Consultancy are happy to announce that we now are producing TRITC-dextran following an improved procedure yielding even brighter fluorescent products. The improved degree of substitution of pure Tetramethylrhodamine dye on high quality dextran will enhance reproducibility and accuracy of in vitro and in vivo experiments.
TRITC dextran are prepared from special dextran fractions by coupling to tetramethylrhodamine B isothiocyanate (mixed isomers). The number of TRITC substituents ranges from 0.001 to 0.008 per glucose unit. At these low degrees of substitution, the charge contribution from the tertiary amino-groups on the rhodamine moiety is negligible. All batches are checked for molecular weight, loss on drying and free TRITC. TRITC-dextrans have now been used for 25 years for studying microcirculation or membrane permeability.
Although the fluorescence intensity varies with pH, in the range pH 4-10 the variations are much less than those with FITC-dextrans (see Fig. in Data file for TRITC-dextran).
